Course Name: Basic Flow Cytometry Refresher
Class Category: Basic. The student should be capable of starting up, running daily QC and interpreting basic sample screening on the Flow Cytometer by the end of the course.
Requirements: Microsoft Office Power Point 2010 or later, computer speakers, email address, Skype address.

Hours: 60-62 hrs of study divided between lessons, reading, quizzes, skype conference and final assignment. Approved by CMLTO (College of Medical Laboratory Technologists of Ontario) as a refresher course.

- Introduction to Flow Cytometry.
- Definition of Flow Cytometry and how it works.
- Types of samples used.
- Fluidics in Flow Cytometry.
- Optics in Flow Cytometry.
- Photo Detectors and Electronics in the Flow.
- Linear and Log amplification.
- Basic Cell separation using Forward Scatter, Side Scatter and Fluorescence.
- Color compensation what it is and why it is important.
- Interpretation and Analysis.
- Some gating strategies.
- Reading list.
- Quiz.
Section B:
- QC for Flow Cytometry and why it is done.
- Linear Verification: set up and reading.
- Flow Cell Alignment: set up and reading.
- Setting and checking the discriminator.
- Setting and checking Voltage alignment and fluorescence alignment.
- Setting up and running color compensation.
- Setting up sample controls.
- Reagent QC.
- Quiz.
Section C:
- Immunophenotyping: what it is, markers or antibodies used.
- What are Cluster Designations (CD): list of some basic CD markers defined.
- Sample setup, types of samples that can be used. li>
- Concept of single marker, or antibody probe and multiple markers used in testing.
- Sample setup.
- Gating strategies.
- Quiz.

- Criteria for diagnosis.
- Basic testing Algorithms.
